چکیده

–Phosphorus is one of the most vital macronutrients required for the growth and development of plants. A large number of microorganisms present in the rhizosphere are known to solubilize and make available the insoluble phosphorus in the available form to the plants. A total of fifty phosphate solublizing bacterial colonies were isolated on the Pikovskaya’s (PKV) agar medium, containing insoluble tri-calcium phosphate (TCP), from repeatedly chemical pesticide used agriculture soil of Dhanbad area. The colonies showing clear halo zones around the bacterial growth were considered as phosphate solubilizers. Out of 50 bacterial isolates, 10 isolates showing highest phosphate solubilisation index (SI) ranged from 1.4 – 3.0 were selected for the further study as qualitative as well as quantitative performance. Among these 10 potent isolates, two strains S2 and S30 showed the maximum phosphate solubilization index of 3.1 and 3.0 in agar plates along with high soluble phosphate production of 373.07 mgL and 368.58 mgL in broth culture, respectively. Isolates S2 and S30 belong to genus Bacillus sp. and Pseudomonas sp. as identified by their morphological and biochemical characteristics, respectively. In all the phosphate solubilizing bacterial isolates, decrease in pH was observed ranging 3.2 to 6.2 from initial pH of 6.8 to 7.2. The decrease in pH of the culture medium there by solublizing the insoluble TCP indicated the production of various organic acids by the culture. Keywords––Phosphate solublizing bacteria, pesticide, soil, Bacillus sp., Pseudomonas sp.

جداسازی باکتریهای دارای آنزیم ارگانوفسفوروس اسید انهیدراز و شناسایی سویه دارای بیشترین فعالیت هیدرولازی

Background and purpose: In Iran, Organ phosphorus pesticides such as chloropyrifos and diazinon are widely used in agriculture. These compounds inhibit activity of cholinesterase in nearly irreversible manner resulting in malfunction of nerve impulse transmission. This result in humans can produce illness or even death. Therefore, the present study aims to isolate various bacterial strains...

Azotobacter chroococcum can improve mineral nutrition of plants through N2 fixation and plant growth promoting capabilities. Fourteen strains of A. chroococcum were isolated from rhizosphere of wheat plants grown in different field conditions around Tabriz, northwest of Iran. In a pot culture experiment with sterile soil, wheat plants (Triticum aestivum cv. Falat) were inoculated with 14 bacter...

Isolation and identification of new beneficial bacterial strains from rhizosphere of Citrus sinensis orchards

The rhizosphere is the area around the root of a plant occupied by a unique population of usefulbacteria known as plant growth promoting rhizobacteria (PGPR). In this study, the isolation andidentification of rhizobacteria from orange (Citrus sinensis) orchards using 16S rRNA gene, as well asbiological and biochemical assays is reported. Analysis of 16S rRNA gene was confirmed bybiological and ...

Specific Genomic Fingerprints of Phosphate Solubilizing Pseudomonas Strains Generated by Box Elements

Primers corresponding to conserved bacterial repetitive of BOX elements were used to show that BOX-DNA sequences are widely distributed in phosphate solubilizing Pseudomonas strains. Phosphate solubilizing Pseudomonas was isolated from oil palm fields (tropical soil) in Malaysia. BOX elements were used to generate genomic fingerprints of a variety of Pseudomonas isolates to identify strains tha...

Effect of organic and inorganic additives on some chemical properties of vermicompost, earthworm’s biomass and reproduction

ABSTRACT-The enrichment of bedding for vermicomposting systems with organic and inorganic additives is a technique which can promote the quality of produced vermicompost. Phosphate rock and fishmeal are two available and low-cost substances which can be provided easily and inoculation of vermicompost with phosphate solubilizing microorganisms (PSMs) can enhance the availability...
